Mice can be primed with a MUC1-derived peptide/poly-L-arginine, adjuvanted and boosted with anti-MUC1 mAbs, and lymphocytes isolated and stimulated ex vivo with the peptide/poly-L-arginine. This proposal is based on our recent demonstration that a single dose of anti-MUC1 mAbs expands CD8+ T cells against MUC1 and boosts the response to a second peptide immunization in MUC1-vaccinated BALB/c mice. We hypothesize that clinical antibodies directed against MUC1 may immunize patients in vivo, resulting in antibody titers that can be boosted ex vivo. Furthermore, we hypothesize that this same protocol will improve the immunogenicity of other tumor antigens as well. To address these hypotheses, we propose the following Specific Aims: Aim 1.
